    How the calculation works

    Concentration equals peptide mass divided by diluent volume.

    Draw volume equals target amount divided by concentration.

    For a U-100 syringe, units equal draw volume multiplied by 100 units per mL. Different syringe sizes change capacity and the displayed scale, not the liquid volume.

    concentration = peptide mass ÷ diluent volume
    draw volume = target amount ÷ concentration
    U-100 units = draw volume × 100

    Worked example

    A 5 mg vial with 2 mL diluent has a concentration of 2.5 mg/mL, or 2,500 mcg/mL. A 250 mcg target equals 0.1 mL, shown as 10 units on a U-100 syringe. The vial contains 20 such draws.

    What is reconstitution?+

    In a laboratory record, reconstitution is the addition of a measured liquid volume to a dry material to produce a solution of calculable concentration.

    Does the diluent volume change the total peptide in the vial?+

    No. It changes concentration, not the total peptide mass entered for the vial.

    Why do results differ between syringe sizes?+

    The calculated liquid volume is the same. The graphic and available markings change with syringe capacity; standard U-100 markings represent 100 units per mL.

    What diluent do the calculations assume?+

    None. The arithmetic is volume-based and does not select or recommend a diluent.
