5-Amino-1MQ is a small-molecule quinolinium compound, not a peptide, and is handled and analysed accordingly. It is supplied as a solid salt. Identity for a small molecule is established by mass spectrometry and, where the specification provides it, by NMR; purity is determined by HPLC with UV detection against a stated method. Discovery and characterisation history
The compound was described in the 2010s during work on inhibitors of nicotinamide N-methyltransferase (NNMT), an enzyme of interest for its role in nicotinamide and methyl-group metabolism. It was developed as a research tool for probing NNMT function in metabolic tissue rather than as a therapeutic candidate, and it has no regulatory status in any jurisdiction.
Mechanisms examined in published work
Published work examines inhibition of NNMT with reported potency values in cell-free and cellular assays, consequent changes in intracellular nicotinamide and NAD+ pools, effects on adipocyte differentiation and lipogenesis in culture, and body-composition and metabolic endpoints in diet-induced obese mouse models. Related work examines NNMT expression in adipose and muscle tissue and its association with metabolic phenotypes.
Where the evidence is strong
NNMT is a genuine and reasonably well-characterised enzyme, and the association between its expression in adipose tissue and metabolic phenotype has been examined by several groups. Inhibition of the enzyme by this chemical class has been demonstrated in vitro with measured potency values. Chemical identity and analytical verification of a defined small molecule are more tractable than for peptides.
Where the evidence is thin or absent
There are no human studies of any kind — not randomised trials, not open-label studies, not pharmacokinetic characterisation. The interventional evidence is confined to cell culture and a small number of mouse experiments, typically with modest group sizes, short durations, and a single obesity model. Independent replication of the mouse body-composition findings is limited. Selectivity against related methyltransferases is incompletely reported in the public literature, which matters for any inhibitor used as a mechanistic probe. Oral bioavailability, tissue distribution and metabolism are not characterised in humans. Every claim about this compound in human physiology is extrapolation from mouse data.
